Engineering 3D osteocyte–tumour microenvironments to study bone metastasis and therapeutic response
Project Reference: #2MPQC-HuECM1Preferred Project Start: Late 2026/Early 2027How do cancer cells communicate with bone cells, and can we recreate these interactions in the laboratory to improve therapy testing? When cancer spreads to bone, tumour cells enter a highly specialised microenvironment containing multiple cell types and a complex extracellular matrix. Osteocytes are embedded throughout this matrix and form an interconnected cellular network that senses and regulates changes within bone. However, their contribution to cancer progression and treatment response remains poorly understood.Conventional two-dimensional …

Prostate cancer transcriptomics (Honours and Master of Philosophy)
At the Australian Prostate Cancer Research Centre QLD, we are interested in the cellular adaptive response processes leading to therapy resistance in advanced prostate cancer.A focus area of our research is studying the transcriptome changes in prostate cancer cell lines, xenograft models and patient samples using RNA sequencing technologies.By integrating our large in-house repository of RNAseq data sets with publicly available studies, this project will further explore the cellular heterogeneity of prostate tumours and the plasticity of cancer cells in …

Investigating differences in downstream signalling mediated by two isoforms of FGFR2 in endometrial cancer
FGFR2 encodes two alternatively spliced isoforms that differ in their ligand binding domain and the combination of tissue specific expression of these isoforms and tissue specific expression of the FGF ligands is the foundation of normal paracrine signalling. Isoform switching from FGFR2b (inclusion of exon 8) to FGFR2c (inclusion of exon 9) occurs in tumorigenesis as it establishes an autocrine loop in epithelial cancer cells. We have previously published a detailed investigation into differences between wildtype FGFR2b and mutant FGFR2b following …

Biological and clinical impact of the association of germline variations in KLK3 (PSA) gene in prostate cancer
Prostate cancer is the most frequently occurring cancer (after skin cancers) in Australian males, and the second most common cause of cancer death. While the 5-year survival rate for localised disease approaches 100%, extra-prostatic invasion results in a poorer prognosis. Kallikreins are serine proteases, which are part of an enzymatic cascade pathway activated in prostate cancer (Lawrence et al 2010). The most well-known member is prostate specific antigen (PSA) or the KLK3 protein, encoded by the Kallikrein 3 (KLK3) gene, …
