Investigating DNA repair mechanisms in aging adult stem cells

When we age the DNA repair systems of our cells become down regulated. This results in reduced DNA repair capacity, enhanced rates of mutation load and may lead to the development of chronic aging-associated diseases including osteoporosis, Alzheimer's and cancer(1). So it is no surprise that genome instability and stem cell exhaustion, which also strongly correlates with the accumulation of DNA damage, are considered hallmarks of aging(2).However, we still lack a clear understanding on how the decrease in DNA repair …

Characterise a novel DNA repair protein as a target for cancer therapies

Data generated in the lab has identified a novel DNA repair protein previously described as a key protein in HSP70/90 complexes. Many pathways of tumourigenesis are mediated by Heat Shock Proteins and HSP70/90 are found significantly upregulated in ovarian cancers. The targeting of HSP70/90 are an emerging therapeutic avenue for the treatment of ovarian cancer. Supporting this, an inhibitor of HSP90 has been shown to sensitise breast cancer cells to PARP inhibitors and paclitaxel. Our preliminary data indicates that this new …

Characterising the role of PARPs in DNA repair and cancer therapy

The genome of our cells is damaged multiple times each day, by various factors including sunlight and reactive oxygen species. In order for the DNA damage response to be efficient, our cells utilise highly coordinated repair pathways that function accurately and rapidly throughout the damaged cell. Cells that do not repair DNA damage correctly will accumulate damage and display increased genomic instability, which is a key hallmark of cancer cells, promoting their survival and rapid growth. DNA repair pathways are …

Curtailing corporate tax aggression through uncertain tax benefits

Over the last five years Australia has adopted numerous measures to address aggressive corporate tax practices. A recent addition to these measures (on the 1st of January 2019) is AASB Interpretation 23 - Uncertainty over Income Tax Treatments, which was developed to clarify the treatment of uncertain tax positions. This interpretation requires entities who produce general purpose financial reports (GPFRs) to disclose uncertain tax benefits (UTB) in the notes to their financial statements. Extant literature suggests that UTB disclosures are …

Identifying AI-amenable tasks in the patient care workflow and matching enabling AI technologies

Contemporary patient care is delivered through complex, multi-step workflows that span triage, assessment, diagnosis, documentation, care coordination, treatment, and follow-up. Many steps in these workflows are repetitive, cognitively burdensome, or prone to delays and errors, and a substantial share of clinicians' time is consumed by tasks that require limited clinical judgement, such as documentation, information retrieval, and administrative coordination. Persistent workforce shortages, rising demand, and clinician burnout have intensified interest in whether some of these tasks can be safely delegated …

Supply chain vulnerabilities and risks in major infrastructure projects: future proofing Australia's construction supply chain

The repercussions of COVID-19 pandemic, Ukraine war and the conflicts in the Middle East has caused global supply chain disruption. Australia is particularly vulnerable to supply chain disruption due to its unique geographic location, construction environment and dwindling manufacturing base.Significant cost overruns, project delays, productivity decline and compromised quality products and facilities have become norm rather than exception. We have a huge demand for major infrastructures including those needed for the Brisbane 2032 Olympic. Future proofing the construction supply chain …

Multimodal AI to simulate medical student competency

The assessment of medical graduate competency is a cornerstone of medical education and a critical safeguard for patient safety. Newly qualified physicians must demonstrate a broad range of skills and knowledge, including diagnostic reasoning, clinical decision-making, communication, procedural skills, and professionalism before independently practicing medicine. Traditional assessment methods often include standardized multiple-choice examinations, objective structured clinical examinations (OSCEs), direct observation of procedural skills (DOPS), and portfolio reviews. While these methods offer valuable insights, they have inherent limitations. Standardized tests may …

Efficient Parameter Estimation for Stochastic Simulations

Stochastic simulation-based models are routinely used in many areas of science to describe inherent randomness in many real-world systems. Applications include the study of particle physics, imaging if black holes, biochemical processes, the migration of animals, and the spread of infectious diseases. To apply these models to interpret data requires statistical methods to estimate model parameters.Unfortunately, standard statistical techniques are not capable of analysing data using these models. This is largely due to the model likelihood, the probability of the …

Development of a Microfluidic Gut-Brain Axis Chip

The gut microbiome refers to the collection of micro-organisms that are living symbiotically in the human or animal gastrointestinal tract (defined as the “microbiota”), their genetic material as well as the surrounding environmental habitat. It is now appreciated that the microbiome plays an important role in human health and diseases. Many neurodegenerative diseases, such as Parkinson's Disease have been linked to dysregulation of the gut microbiota. However, it is difficult to study gut-brain axis using animal models due to inter-species …

Making the most of many models

In the age of Big Data, machine learning methods, and modern statistics the adage "all models are wrong but some are useful" has never been so true. This project will investigate data science approaches where more than one model makes sense for the data. Is it better to choose a single model or is there something to be gained from multiple models? This project will look at variable selection methods, penalised regression, Bayesian model averaging and conformal prediction. The research has …
