Shape and topology optimisation enable the design of material layouts to give improved mechanical or other properties subject to physical constraints. These computational techniques are relevant to a wide range of industrial applications and have a substantial mathematical foundation. The thesis aims to advance and improve the accessibility of computational techniques for level-set topology optimisation and apply the developed techniques to the design of extreme piezoelectric metamaterials. The thesis provides theoretical contributions to optimisation algorithms, efficient numerical methods, and shape calculus. It also provides new insights into piezoelectric metamaterial design for engineering applications.

Optimising Remote Sensing to Spatially Predict Above-ground Biomass and Soil Organic Carbon at Field Scale in Subtropical Grasslands

Grasslands are important for storing carbon, but measuring soil organic carbon (SOC) is difficult due to high variability and costly sampling. This research evaluated whether satellite remote sensing, combined with field observations, can improve estimation of above-ground biomass (AGB) and SOC in subtropical grasslands of Queensland. Results showed that integrating satellite data with visual pasture assessment and stratification approach improves SOC prediction and reduces uncertainty. Satellite-only models provided moderate accuracy and can support large-scale monitoring. Overall, this study demonstrates a cost-effective approach for carbon estimation and supports improved sampling strategies for carbon-credit projects and sustainable land management.

This thesis addressed limitations in the use of Nicotiana benthamiana for forward genetics arising from its large, polyploid genome by developing a high-throughput trait mapping and gene discovery workflow. Using two genetically and phenotypically distinct accessions, LAB and QLD, the study investigated their contrasting responses to anthocyanin induction by integrating genetic crosses, bulk segregant analysis, genome sequencing and transcriptomics. The detrimental LAB response was mapped to a genomic region containing a leucoanthocyanidin dioxygenase gene, which was functionally validated by complementation. This work demonstrates that forward genetics can be applied effectively in N. benthamiana, enabling gene discovery despite its genomic complexity.

Bioactive molecules such as proteins and heavy metal ions are of great concern due to their effects on human health, driving the need for sensitive and reliable analytical detection methods. This research focuses on developing novel sensing materials and platforms for detecting environmental pollutants and proteins using colourimetric, fluorescence, and surface-enhanced Raman spectroscopy (SERS) techniques. A new diketopyrrolopyrrole dye (DPPT-PhSMe) and an isoindigo-based sensor (II-MT) were developed for selective and accurate Cr(VI) detection in water. Additionally, a label-free SERS paper sensor detected the cancer biomarker S100P with high sensitivity, demonstrating high potential for early cancer diagnosis.

What is the Role of Ghrelin in Equine Appetite and Insulin Dysregulation?

This thesis investigated the relationship between appetite and the metabolic disorder insulin dysregulation, which can lead to a painful disease of the hoof in horses and ponies. The research undertaken discovered individual animal variation in appetite that could predispose to disease risk; and identified associations between the appetite hormone ghrelin and breed, measures of hunger, and of satiation. Animals diagnosed with insulin dysregulation had lower circulating ghrelin concentrations than their healthy counterparts, indicating that appetite regulation may be an important factor in insulin dysregulation in horses and ponies.

This study uncovered dramatically different survival strategies to water deprivation within the same species of an Australian native plant, Nicotiana benthamiana. One strain, from central Australia, has evolved the drought escape strategy of early vigour and short life cycle, with little responsiveness to water scarcity. In contrast, an accession from Queensland, responds to water scarcity by slowing its growth rate, inhibiting flowering, and altering its cuticular wax composition under the control of the previously unrecognised transcription factor (NbMYB6) to minimise water loss. This prolongs the plant’s capacity to survive without water until follow-up rainfall supports further growth and rapid flowering.

Modern slavery affects millions of people and is often hidden within corporate supply chains. Modern slavery laws require companies to publish statements on the actions they take to address these risks, but the growing number of statements makes manual review difficult and limits accountability. This thesis develops and applies artificial intelligence to analyse, benchmark, and strengthen disclosure assessment. It creates large annotated datasets, fine-tunes and benchmarks language models, and builds tools for compliance checking, cross-jurisdictional analysis, and quality assessment. The findings show how AI can support policymakers, regulators, civil society, and investors through more scalable, transparent, and evidence-based reviews.

This thesis explores high-performance flexible thermoelectric materials, focusing on n-type Ag2Se and Ag2Te thin films for near-room-temperature applications. A hybrid synthesis approach combining physical vapor deposition with wet-chemical selenization/tellurization was developed to optimize microstructures. Key strategies, including Indium doping in Ag2Se and controlling residual Ag in Ag2Te, effectively decoupled the trade-off between carrier concentration and mobility. These methods achieved ultrahigh electrical conductivity and a record-breaking power factor in flexible films. Finally, integrated flexible thermoelectric generators demonstrated superior power density and mechanical durability, highlighting the potential of these materials for sustainable wearable electronics and self-powered sensors.

This thesis introduces a sustainable material-to-cell engineering framework to accelerate the commercialization of aqueous zinc-ion batteries (AZIBs) for grid-scale energy storage. To overcome foundational degradation modes, three interconnected pillars were developed: (1) an ultrathin mixed-conducting Ti3C2Tx MXene-carbon fibre protector to regulate interfacial electric fields and suppress out-of-plane dendrites; (2) an industrially scalable, materials-lean 3D anode fabricated via aerosol cold-spraying to maximize active mass utilization; and (3) an expanded, pillared bilayered V2O5 nanostrand cathode processed with water-borne, fluorine-free binders to eliminate manufacturing toxicity. This PhD thesis develops advanced statistical methods within a Bayesian design framework to enable deeper parts of reefs to be efficiently and effectively monitored. This research addresses three main objectives: (1) Develop model-robust Bayesian design methods to optimise sampling under model uncertainty; (2) Advance Bayesian design in spatio-temporal settings via variational approximations for efficient computation; and (3) Develop a framework to leverage information from data-rich reefs to monitor data-poor reefs. Collectively, these contributions to the Bayesian design literature enable robust and efficient designs to be found and used for sustainable monitoring and management of our deeper reef ecosystems.

This thesis presents statistical tools to improve the prediction of animal movement. Animal movement was modelled using statistical models that captured periodic behavioural patterns, as well as deep learning, both of which can generate individual-based simulations. We also present a conceptual framework for predictive movement ecology, where these models can be used for a range of scientific and conservation management purposes. The concepts and models could be used for a range of species, but here were applied to tracking data of introduced water buffalo and feral cattle in northern Australia, thereby providing valuable information for land managers.

This research focused on developing a comprehensive macroscopic model for describing the moisture-induced swelling stresses within radiata pine glue-laminated engineered wood products. An image-based mesh generation algorithm was created to generate a three-dimensional virtual representation of the product that captures the heterogeneous structure of timber. This representation was then used to simulate the moisture migration and heat transfer processes throughout the product and a transfer coefficient for the glue line was determined. The swelling stresses were then computed and points of failure identified. The model was then demonstrated by predicting delamination in a variety of climates and wetting scenarios.

Utilizing magnetron sputtering and ex-situ annealing, the research achieves record-high power factors (S2s) in n-type Mg3Bi2and p-type Mg3(Sb,Bi)2 thin films. These films demonstrate exceptional mechanical durability, retaining performance over 1,000 bending cycles. By optimizing bismuth content, p-n junction devices were fabricated, proving effective for wearable energy harvesting. Additionally, an innovative melt-synthesis strategy enhances bulk production scalability. Overall, the work establishes a robust framework for integrating high-performance magnesium-based thermoelectrics into the next generation of flexible electronics and commercial power systems.

Although plastic use plays a crucial role in food packaging applications, these plastics pose significant concern due to the persistence of plastics in the environment and their subsequent accumulation. By developing a polymer from brown algal species, there is potential offset plastics as food covering agents, whilst also utilising bioactive components within brown algae to increase food safety and stability. This thesis explores an integrated design approach to develop brown algal derived polymers, from polysaccharide extraction, to film formation, and subsequent silicate functionalisation procedures to develop polymers aimed at offsetting plastic use for existing food covering applications.

Predictive regression models are often used to forecast future possibilities of a given finance variable. For that, we rely on statistical inference: estimation and hypothesis testing. Inaccurate estimation results make inaccurate inference for a scientific question. So, it is important to develop methodologies to reduce the bias in the estimation providing a sounder basis for statistical inference. Hence, the contribution of this research is to deliver more reliable estimators in terms of bias and the level of persistence of the predictor variable, and to develop a corresponding inferential framework with time-series and longitudinal data.

Comparative functional analysis of FT, TFL1 and FD homologs reveals conserved flowering pathways in avocado, macadamia and mango

This thesis investigates the flowering regulators FT, TFL1, and FD in avocado, macadamia, and mango, three economically important perennial fruit crops with poorly understood flowering behaviour. Homologs were identified, characterised, and analysed using phylogenetics, expression profiling, promoter analysis, protein interaction studies, and Arabidopsis complementation assays. Results showed that the FT–FD–TFL1 flowering module is broadly conserved, while gene duplication and divergence have led to species-specific functional specialisation. Key floral inducer candidates were identified, including PaFT in avocado, MiFT5 in macadamia, and MaFT4 in mango. These findings improve understanding of flowering regulation and provide future crop improvement strategy potential targets.

Battery performance using high-purity, nickel-based cathode active materials is impaired when made from recycled feedstocks containing copper, iron, and other contaminants introduced during the recycling process. This work produces a reduced-cost pathway for cathode active material synthesis able to exclude much of the problematic copper which resulted in cell performance competitive with commercial benchmarks. It demonstrates a vertically-integrated refining and remanufacturing process able to create commercially viable cathode active materials from recycled lithium-ion batteries.

This thesis explores a new class of glass made from small molecules that assemble through dynamic interactions, rather than the strong chemical bonds found in traditional glass. By designing different supramolecular glass systems, this research demonstrates materials that are transparent, processable, adhesive, and responsive to moisture. These glasses can be shaped into films, fibres, and coatings, and can also support light emission and self healing behaviour. The findings provide new understanding of how molecular assembly can be used to create functional glass materials for future optical, coating, and advanced manufacturing applications.

The models developed in this thesis were applied to avocado fruit growth data collected in Western Australia over three consecutive growing seasons. This was one of the first studies to quantitatively characterise avocado fruit growth under Australian conditions while accounting for variability between trees, orchard position, and growing season.

A key challenge is developing durable and efficient catalysts for the oxygen evolution reaction. The thesis investigated iridium oxide (IrO2), a leading catalyst, and enhanced its performance by incorporating small amounts of barium, copper, and lead. These modified materials demonstrated significantly greater stability and activity in acidic, neutral, and alkaline environments. Mathematical equations that describe mass transport through heterogeneous materials are highly complex and difficult to solve. This thesis details the development of some novel computational and analytical methods for solving such equations in an accurate and efficient way. In real-world scenarios, such as chemical contamination in aquifers, it is common for heterogeneity to occur over both small and large length scales. Accordingly, this thesis develops a new computational solution method for simultaneously obtaining solutions at both length scales and a new analytical solution method for upscaling certain parameters from small to large length scales.

Citation

Protecting Antarctica's ecosystems in a changing world requires detailed and timely knowledge of the continent’s biodiversity. Predictive spatial ecological models can enhance the value of available and emerging Antarctic biological data but are currently underutilised on the continent. This thesis aims to advance the prediction of Antarctic species distributions in space and time to improve the conservation of Antarctica’s life on land. I achieve this by addressing barriers to the widespread uptake of predictive models in Antarctica.

Advanced Nanomaterials Enabling Clean Manufacturing via Sustainable and Photoswitchable Photocatalysis

This thesis presents the development of nanomaterials that enable solar-driven chemical reactions to occur through visible light photocatalysis. Copper and gold based nanocatalysts, including Au–Co alloy nanoparticles, were engineered offering a greener and environmentally friendly approach to chemical synthesis. A novel organic thiol-based photocatalytic reaction was also reported for the first time. The findings contribute to next generation photocatalysis and sustainable technologies, offering viable solutions for green manufacturing and energy efficient chemical production. The work aligns with global innovation goals in nanocatalysis, clean energy utilisation, and the application of advanced materials for a low carbon future.

Australian wine growers have faced mounting challenges from climate change, water scarcity, and volatile markets. This research explores how to help balance the environmental and economic elements of sustainability; using industry data and expert knowledge, the research develops statistical models to understand how different vineyard practices effect viticultural outcomes. Two contrasting Australian wine regions—the cool coastal Coonawarra and warm inland Riverland— were examined to reveal how local climate and geography influence farming decisions. The resulting framework provides growers and policymakers with useful tools to understand key trade-offs and make sustainable choices despite uncertainty.

Plants are essential to life on Earth, providing food and maintaining the global Carbon cycle. Mathematical models can predict and optimise plant growth, but their usage relies upon model behaviour being realistic and good estimation of their parameter values. Using a model for Carbon/Nitrogen allocation in plants as a case study, this thesis identifies non-physical and chaotic behaviours under some parameter regimes and proposes a modification to improve stability. Using statistical techniques, parameter identifiability is assessed across different data types. The results highlight which measurements are most informative and how model analysis can guide experimental design when data are limited.

This thesis explores the development of new “hybrid” drug molecules designed to better treat diseases linked to inflammation and oxidative stress, such as cancer. By chemically combining anti-inflammatory steroids with redox-active nitroxides, this research developed novel drug candidates that both regulate inflammation and disrupt cancer cell survival, with studies demonstrating not only their specificity towards malignant cells, but also their synergistic enhancement of existing chemotherapy drugs. Overall, this research provides a foundation for designing smarter, more effective treatments that target multiple disease pathways at once.

Mining for Fusarium Wilt Susceptibility Gene Targets in Banana for Editing and Disease Resistance

Bananas are one of the world’s most valuable agronomic commodities, yet production is greatly constrained by disease. Fusarium oxysporum f. sp. cubense (Foc) has had a devastating impact on the banana industry. This study aimed to engineer resistance to the Foc variant subtropical race 4 (STR4) in the cultivar Lady finger by targeting putative disease susceptibility genes using CRISPR/Cas9. In parallel, transcriptomic analyses of this pathosystem were conducted to identify additional susceptibility gene candidates. This work is the first report of gene editing in Lady finger and provides resources to support future disease resistance efforts in this important crop.

Fabricating Silver Microstructures on Zinc Oxide Thin Films through Laser Driven Photodeposition

This research developed and tested a novel instrument for conducting laser-based photodeposition to fabricate silver microstructures, providing significant improvements in the reliability and reproducibility of the initiating light in photodeposition reactions. The precision increase allowed the study of controllable optical properties during these reactions, highlighting a fluence threshold and providing early evidence for an interplay between optical and chemical properties in controlling the morphology of fabricated structures. Results demonstrated the benefits and limitations of laser-based systems, with recommendations made to integrate the learnings from both into future research, including for use with non-Gaussian beam shapes.

This thesis investigates how extreme events and the policies responding to them reshape people’s benefits from nature. It shows that shocks such as COVID-19 and climate mitigation policies can unintentionally reduce access to green spaces and shift environmental pressures onto vulnerable countries. Using conceptual, empirical, and global modelling approaches, the research demonstrates how shocks and policy responses alter ecosystem services across cities and nations, and highlights the need for better planning and international cooperation to ensure fair and sustainable outcomes.

Nature holds secrets to plethora of properties including strength, toughness, and adaptability. My PhD explored three remarkable structural inspirations: the sharp, strong leaf tip of Agave striata, the adhesive micro-hairs on a fly’s leg, and fibers of the protective silk cocoon. By studying their in-depth unique architectures and mechanical properties, I developed bioinspired platforms for advanced materials microneedles, nano 3D printed nano-arrays for smart adhesives, and fibrous meshes for water purification and sports textiles. These designs combine lightweight strength, resilience, and sustainability, offering solutions for healthcare, environmental technologies, and everyday applications.

Interfacial and Electrolyte Engineering for High-Performance Aqueous Zn Metal Batteries

Aqueous zinc batteries are a safe and low-cost option for large-scale energy storage, but their performance is limited by instability of the zinc metal electrode during repeated charging and discharging. This thesis develops new electrolyte additives and thin protective coatings to make zinc electrodes more stable and reliable. The research shows that regulating ion behaviour at the zinc surface can suppress unwanted reactions, prevent uneven metal growth, and greatly extend battery lifetime. These strategies improve battery efficiency, durability, and safety, offering practical solutions for future sustainable energy storage technologies.
